Abstract

Comparison of Monoclonal and Polyclonal Saline-Reactive Rh-Reagents by Erwin Strobel

To examine whether there are still any advantages of polyclonal saline-reactive reagents for Rh-typing in comparison to monoclonal reagents, ten pairs of monoclonal and polyclonal reagents - both of the same specificity and from the same manufacturer - were tested. All monoclonal reagents had higher titers with untreated red blood cells (RBCs) (carrying the corresponding antigen) than the polyclonal reagents. Several of the polyclonal but only few of the monoclonal reagents showed false positive reactions with RBCs (without the corresponding antigen) coated with incomplete antibodies of another specificity. No false negative reactions caused by blocking phenomena were seen. Since no advantages of polyclonal saline-reactive Rh-reagents were found in our study, we prefer the use of saline-reactive monoclonal Rh-reagents of the lgM-type.
